I have a bunch of western blot results of mitochondrial complexes which i obtained using 2D gels followed by western

PI wants me to do densitometry on them for quantitation

Now issue is - i don't have anything to compare it with. I have a single blot of complex I ... and even the molecular ladder does not show up [we use a licor imaging system to develop blots] as i run a blue native gel & the ladder gets stained and never shows up

i can get intensity values for my band but how does that ever help me? i can't compare it with anything. is there a way i can know the quantity of my protein expressed [as i know how much protein i had loaded before]

you can use a biotinylated ladder and process with a biotin-avidin hrp or ap (depending on which enzyme you use for your western blots).
