StrepTag Missing Amino Acid

Hello,

I recently cloned a lot of variants based off of a friends primer designs. The tag we used was a classic strep tag WSHPQFEK. He designed the primers so they cut off the lysine. The tag we present is WSHPQFE*

I was wondering if its worth it to mutate the lysine in there or if this will work. If I do add the lysine, it will be a ton of extra work.

Depends on what you will use the tag for. If I remember correctly streptavidin binds the HPQ residues, so you should be OK, without the Lys. However, if you will be using any anti-streptag mAbs, I would not be so sure it would work.
