Culture of Whole blood in vitro

Hello,

I am involved in a project where we are interested in the use of whole blood. Usually I transfer the whole blood from EDTA tubes to flasks with filter caps (for gas exchange). I dilute it with 1:1 RPMI.  Upon incubation of whole blood, genes related to hypoxia are coming up. Do you think whole blood need extra oxygen supplement for in vitro culturing? Is there a standard protocol to culture blood in vitro?
