Hello all,

Just a quick question. I know a lot of companies and individuals design their siRNAs with 3' dTdT overhangs. My question is, are these dTdTs added on regardless of the gene/RNA sequence, or do the dTdT have to be inherent within the genetic sequence to start with?

E.g. If I had a 19-mer sequence of GCCTACTGCTTAGCTGACT and there was no complimentary dTdT occurring naturally at the end, is it OK to just add it on? Does the dTdT play a role in any way other than in protecting against exonuclease activity?

I hope this makes sense, not sure I've explained myself very well!
