Hey guys,
I'm currently studying Adv. Higher Biology and am in the process of planning my investigation.
I am planning on using the title "Discuss the effect of temperature on transformation of E.coli K12."
My basic method is going to include making the bacteria competent and then somehow obtaining cephalexin resistant plasmids and transforming the E.coli.
However, we get to my main question: How do I obtain a killing curve for cephalexin. If somebody to tell me a general method then that would eb great. I have a basic idea: grow the bacteria, plate them in conditions with differing antibiotic concentrations and then noting the number that survive. Couls any of you guys give a more detailed plan?
Also, if any of you know where to get cephalexin resistant plasmids or any other antibiotic resistant plasmids then if you could let me know then that would be awesome!
