Hello.
I'm trying to overexpress a protein (wip1) in HeLa cells, using cDNA.
cDNA I'm getting comes in pCMV-sport6 vector, which is a mammalian expression vector.

I'm wondering if direct transient transfection of HeLa cells with the pCMV-sport6 with my cDNA would do the overexpression.

Can anyone help?

It should work, the CMV is a viral promoter designed for over-expression of proteins running off the promoter.
