Any suggestions for doing a co-IP for 2 proteins that are associated with each other at the cell membrane. If you disrupt the cell membrane with CHAPS or Brij buffer, will you disrupt interaction of 2 membrane associated proteins that are interacting at the membrane? Would using sonication instead of detergent disrupt protein-protein interaction or complexes? We are trying to find a balance between dirupting cell membrane to open up cells for IP and preserving interaction between 2-membrane associated proteins. Any suggestions are most welcome.
