Does anyone here have experience on doing NCFC using adult mice? This assay is basically growing neurospheres in collagen. I've tried different digestion methods, (i.e. trypin at different concentrations, papain) and seeding densities but no luck. I've always run the "traditional" free-floating neurosphere assay in parallel and they were just growing fine. All I've found out there in the literature use embryonic or postnatal mice which don't require digestion at all. Can anybody help?
