Hi~
can anyone give comment on how to isolate Yersinia enterocolitica from pork meat?
i've been working on it for months and the isolation were poor.

i use 3 enrichment broths

yersinia selective enrichment broth according to OSSMER, YSEO (Merck) 25C, 1d
irgasan-ticarcillin-cholate broth, ITC (Fluka)25C, 2d
phospahte buffer saline, PBS (Sigma), 4C, 3 weeks

then

plating onto yersinia selective agar, CIN (Oxoid) and Salmonella Shigella Deoxycholate, SSDC agar
25C, 18-24h

so far i only get few positive results using PBS enrichment + CIN agar plating but 100% negative for YSEO and ITC enrichment methods.
PCR screening results showed positive but negative when culturing.
is there any way to increase the isolation efficiency?
