I have a query regarding analysis of real time PCR results. I am comparing gene expression levels of two gene isoforms with gapdh as internal reference. i have to compare relative gene expression levels in drug treated and untreated samples. Since delta Ct (Ct of target-Ct of reference) is inversely proportional to mRNA levels, i inverse delta Ct and compare between two isoforms in the samples. but, I have not come across any paper that use 1/delta Ct to compare expression levels. Is it correct to compare that way? In my case, fold change by delta delta Ct do not provide adequate explanations.
