Voltage clamp for AMPAR and NMDAR

Could anyone help me figure out why we usually set holding potential at about -70mV and +40mV while recording AMPAR and NMDAR-mediated synapse currents, respectively?

and why usually NMDAR shows upward deflection, yet AMPAR shows downward? what do upward and downward mean?
