I want to tranfer a plasmid 12kd to DH5a(E.coli) and then get enough plasmid DNA to transform the agrobacterium LBA4404 ,but I did that for five times ,I only got one clone in two of which,and I got DNA from the clone then tranformed the LBA4404 in the way of freeze-thaw, but got nothing.Who can tell me why and how to solve the difficuty,thanks!                                            naizhong
