Hi, everyone,

I am just doing P1 transduction. I need to purify the pCP20 plasmid to remove the Kan+ cassette in my receipt cells. But I have tried twice from different strains containing pCP20 (BT340, BW1627) and I didn't get any plasmids. Both of the strains are new and have Amp+ resistance.

Is there anybody know how to mini prep the pCP20? is there any specific requirement for the purification?
