I don't really know if I am in the right topic. I study the effect of an up- or down-regulation of a gene on proteasome activity in HeLa cells. In untreated cells, siRNA or plasmide of my gene decrease proteasome activity. It seems so weird to have the same effect with plasmide and siRNA... Have you ever seen such an effect? On the other hand, when I treat HeLa cells with a proteasome inhibitor, I decrease proteasome activity with siRNA whereas I increase it with plasmide (all compared to a control siRNA, and I checked up- and down-regulation of my gene by RT-PCR).
Has anyone ever seen a gene that, when up- or down-regulated, has the same effect on a function? I was unable to find the answer in literature. I would just like to have an example, first to know if this mechanism is possible, and to study it more deeply.
