Hi,
I am trying to make standard curve for real-time PCR. I have amplified the cDNA using gene specific primers and then run it on gel. Now I am about to cut the specific fragment in gel, purify and quantify it. It seems very complicated and sensitive.

Can I use the cDNA (got by using OligoDT) for making standard curve dilution for real-time PCR?

What kind of quantification are you planning to do? Absolute or relative?

I want to see the gene expression at the end. So I am not bound to any, any of them would be nice.
