Hi all

I am designing primers for pcr amplification of 2.3kb insert and I need to add salI primers at the 5' end of both the primers.Can anyone please suggest about the appropriate bases that I need to incorporate before the restriction site at the 5' end?

I'm new with plant transformation and rt-pcr. Please help me find  promoter and primer sequences for Arabidopsis and tomato plant. And please let me know, if there are any factors that I have to consider in order to do rt-pcr.
