Hi All ,
I want to stain helminth eggs but the problem is it has a lipid layer which is highly impermeable. Does any one have any idea about improving the permeability of lipid layer ( espeacially ascaroside layer)? like treating with xylene, or sonication or electoporation?

Cold Aceton treatment should fix the cells and dissolve away lipids along the way.
