Hi all,

I've been working for the last month to generate stable HeLa cell lines expressing various tagged histone constructs, but no matter what I try doing by the time I've selected colonies (12-20 for each construct) none express the proteins.

I've tested the antibiotic concentrations and have checked the original transfection efficiency. Other labs have used these constructs to establish stable cell lines so I have no clue what is going on.

Could you provide some more informations, like how you try to establish the cells, how long you select, what selection you use etc?
