Hi,

How important is it to wash with PBS after peroxidase block in IHC.  I added blocking buffer to my slodes after 30min peroxidase block and then realised after 10 mins that I had to wash with PBS first. I then removed the blocking buffer and washed with PBS. Should it be alright?

the peroxide block is to kill off any endogenous peroxidase activity. since you are not adding peroxidase until later in the procedure, the lack of washing before (protein?) blocking should cause no significant problem.
