Does anyone have the same problem as I have? I digested both MSCV and MigrI with HpaI, which should be a unique site in the multiple cloning sites. However, when single digestion by HpaI, I always get three bands, one around 1.7kb, one largest band migrating at the same level as the supercoil vector, and the other migrating a little bit faster than the largest one. Does anyone have any suggestions? Actually, when I have that digestion problem with Migr I, I changed it to MSCVpuro from Clontech, and got the same problem.
