I am wondering if anyone has any thoughts on whether using a RNA primer for standard PCR wouldn't work?? I am concerned about the heat stability of the RNA primers and I can't seem to find any literature where others have used an RNA primer in a PCR.
I want to use a RNA primer because I need to get rid of all excess primer so I figured I could use a RNase after the PCR to remove. I have tried many other ways to get rid of excess primer (i.e. biotin/streptavidin, columns, gels etc), however, the technique I am trying to work up is extremely sensitive so none of these have worked well enough and there is always carry over.
