Hi, I want to fix HEK293-YFP cells to observed them in confocal microscopy. I also need to use a DAPI staining to observe the nuclei, so I wonder if I could follow a normal immunofluorescence protocol, fixin cells with 4% PFA and permebealize with 0,1 % Triton x100.

You should be able to I think. We typically fix, permeabilize, do any immuno/staining procedures, then add vectashield with DAPI. This of course has wash steps between each of those.
