Iam startig to wrk in a new project,my first task is to do PCR from lyophilized varicella vaccine (Oka strain)
Should I do direct PCR or DNA extraction and which protocol should I use

DNA extraction should give better (as in more consistent) results.  A standard phenol-chloroform preparation should work well.
