I was ask to optimize the promoter that I am using in my vectors. Any free guide or step by step manual to follow? I really don't know what to do!

What do you mean optimize? Is it mammalian, bacterial or other?
For what purpose do you need it?
Is it not expressed well in experiments?

Please be more specific.
