hello, I have been doing some DAB staining and think that something went wrong with the reaction. Our current protocol describes the DAB reaction only taking approximately 5-10 min to occur and it took about an hour the last time i tried it- what could have gone wrong? I also noticed after DAB staining it was extremely hard to see structural landmarks(like the hippocampus and the internal and external capsule)unlike after our regular immunofluorescence protocol where structural landmarks are very easy to identify. Im attempting to count and label cells in the amygdala and its difficult because i cant see structural landmarks. Does anyone have any suggestions?
