Hi!

I'm trying to clone for the first time a new gene in a fish specie by homology cloning. Comparing with other organisms I could see that the region I'm trying to clone is highly repetitive and codify to a conserved and characteristic domain of the protein.

So, I wanted to know if someone knows some technique that improve the PCR amplification in repetitive regions? I have heard something like add glycerol in the reaction...
