I am looking for a step-by-step protocol describing how to extract protein from freeze-stored fat and muscle tissues, which method would be appropriate for detecting PPAR gamma by immunoblotting. And How much protein is typically extracted from a fat tissue?

I am currently developing a fluorescent immunohistochemical protocol for the detection of both PPAR gamma and C/EBP alpha in bovine muscle tissue. Preliminary results are promising using commercially available primary antibodies. Once this is done I will be screening for the two transcription factors using Western Blots and will therefore need to perform protein purifications of both muscle and fat tissues.
