I am looking for a step-by-step protocol describing how to extract protein from freeze-stored fat and muscle tissues, which method would be appropriate for detecting PPAR gamma by immunoblotting. And How much protein is typically extracted from a fat tissue?
Thank you for your kindness in advance.

I am currently developing a fluorescent immunohistochemical protocol for the detection of both PPAR gamma and C/EBP alpha in bovine muscle tissue. Preliminary results are promising using commercially available primary antibodies. Once this is done I will be screening for the two transcription factors using Western Blots and will therefore need to perform protein purifications of both muscle and fat tissues. If you wish you can contact me privately and I can provide the protcols via email to you directly.
