Hi all,
I would like to know if histone H3 migrates at a different height in tris tricine gels... In other words, is it possible that H3 appears in WB as a 25 KDa band?

Histones are very positively charged. Histone H2B and H3 runs between 20-25 kDa in my gels (real MW is ~ 15kDa). Also, many pre-stained protein markers are not very accurate for MW detection. If you are not sure you are detecting histone, run some purified histone in your gel. Good luck!
