I have encontered Y2H problems. I used Invitrogen method. First, I transformed bait construct(pdBleu::TargetGene)into yeast Mav203 strain. For negative control I replaced bait construct with water. Later I found both transformation produced colonies. Then I extracted plasmid from ten colonies, but it turned out that none of them has target gene. Did my problem occured in transformation step? Why colonies grew in negative control? Look forward to anyone's advices. Many thanks!!
