I need to co-stain two antibodies. One works perfectly with citrate buffer antigen retrieval (AR); the other one is a fluorophore-conjugated primary antibody against GFP that works perfectly without AR. To co-stain them, I need to do AR for one of the antibodies to work. I first stain with the directly conjugated GFP antibody. I then fix it to the tissue with FA. Then I do AR. However, the process destroys the fluorophore of the GFP antibody (I think), eliminating the staining of that antibody. Any suggestions how I can perform antigen retrieval without destoying the fluorophore? (NB. I think I must do the GFP staining before I perform AR because the GFP ab epitope seems to be destroyed by the AR)
