I used to extract plasmid with conventional method and after running on gel I could see the linear and circular bands intensity were much stronger than the supercoiled band.
These days I use PureLink kit from Invitrogen and the linear band is almost invisible but the supercoiled band intensity is very strong. surprisingly my transfection efficiency is also much higher. Is the higher amount of supercoiled plasmid the reason?
