I want to check the status of acetylated histones H3 and H4 in westen blot. for this do is it neccessary to extract histones or Can I run western without this?

Though, I haven't done that before, i would recommed you perform Acid-Urea gel which cleanly separated histones, histone variants and differently modified histone isoforms (such as acetylated and phosphorylated histones) on the basis of differences in their charge. You can refer to "extraction, purification and analysis of histone" (Nature Protocol) .
