Hi all,

Just a doubt. When HpaI cuts DNA of interest, it generates a blunt end. I just want to check if the blunt end has a phosphate group in order for ligation to occur with a vector (also cut with HpaI)?

Yes it does...watch out for recircularization during ligation.
