I'm looking for reasons on why I would get accelerated growth of E. coli ML35. My supervisor prepared a culture of OD600 = 1. So initial concentration should be 10^8 cfu/mL. Diluted to 10^6 by using 4.95 mL of MHII (growing medium we're using with all cultures) and 50 uL of 10^8. Diluted to 10^5 by using 4.5 mL of MHII and 500 uL of 10^7.
We record absorbance values in increments of 30 minutes. On average, our cultures don't start becoming cloudy until 3 or 4 hours. Our most recent MIC assay took off within 1 to 2 hours. The only extra time that the microplate wasn't in the spec was probably for about a half hour so that doesn't seem likely. Anybody else have any ideas?
