Hi all,

I am trying to make doubly-fluorescent mammalian cells. I now have stably fluorescent cells with one of my constructs (resistant to G418), but then I realized that the second one is also in a vector that has G418 resistance and that probably won't work. So I want to put the second construct into a vector with puromycin resistance.

Does anyone have a recommendation for a good puromycin-resistant vector with a decently large MCS? I don't want any fluorescent or other protein tags. The first vector I used was pEGFP-C1, which has a CMV promoter. Does it matter which promoter is in the puromycin vector?

Thanks!
