HI,

I'm new to protein extraction.  I had extracted proetin from cell lines using ripa buffer.  Now I need to extract proetins from tumor tissues and soem protocols recommend Urea buffer. What is the difference and is it alright to extract protein from tumor tissues using ripa?

i think the difference is the solubilization of your proteins!
I used ripa buffer to solubilize one membrane protein! when i used buffer with urea, i couldnt solubilize better and see by western blot.
also, im not sure, but it may depends on what you want to do as experiment after.
