MTT Assay Sensitivity

Hi,

I just have a quick question; I've recently started doing MTT assay and I'm currently dissolving the MTT crystals in DMSO and leaving it on a shaker at 37oC for ~15 minutes. I'm just wondering if there would be any drastic changes to sensitivity of the assay between different samples if it was left overnight or for a longer period of time.

yes it would be, because we are talking about cells. do you plate your cells in 96-well plate?

No, they're done in a 24 well plate at the moment.
