Hi,

I'm looking to do histology on mouse adipose tissue but it's proving quite difficult to cut the sections. My current protocol involves putting the sections in 10% formalin for 24 hours, transferring them to PBS and then into 70% ethanol before processing as normal. However the sections are too soft to cut.

Does anyone have a better protocol for processing adipose tissue?

Do you embed the tissue in paraffin? I fix overnight in 4% Pfa, then PBS wash until paraffin. Works fine for me.
