Hi

I have a transgenic NOD mouse. We are trying to breed one of the positive transgenic NOD with a NOD SCID to generate a NOD SCID transgenic. This might be really silly but are the pups going to be still immunodeficient? If the transgene can be transferred to the NOD SCID background, why can't they get the normal immune development traits? My transgene has nothing to do with immune system and it is expressed only in the pancreas.

What? Lol, so why exactly are you using NOD and NOD SCID mice? A more typical (and immunocompetent) strain to transfer a gene onto would be a C57BL x 129 outcross or something

if U r making a NOD SCID transgenic then why should the mouse be immunocompetent?

OK maybe I'm a little confused by the wording in the op. Are you saying that you want to produce pups with or without a functional immune system?

We use the NOD mouse because we work on type 1 diabetes and it is the mouse model for autoimmune diabetes   Also I need an immune deficient NOD SCID transgenic line in the end. I finally got the answer. My assumption was right. The pups I have would be heterozygous with a normal immune system and then I have to do brother-sister matings to generate homozygous immune deficient NOD SCID transgenic mice

Because I am crossing a normal NOD SCID mouse with a transgenic immune competent NOD mouse to generate the NOD-SCID transgenic line.
