I have been doing transformation by the electroporation method for my cloning and I use a ligated plasmid for transformation. however i have never been successful in the cloning. Apart from trouble shooting other areas, I was wondering if the method of transformation could have a role in this. could chemical method of transformation yield the desired result when i am trying to clone with a circularised plasmid as compared to a linear plasmid? Is there a difference between electroporation and the chemical method of transformation apart from the yield?
