HI,

I need to design primers with restriction site to clone the entire reading frame of my gene of interest into a vector.  I understand I have to add a kozak sequence before the start codon.  I know the function of the kzak sequence, but what specific sequence is added?  Is it GTC?

I use ACCATGG where the ATG is the start codon.
