I would like to generate mouse chimaeric for B cell (or CD4 T cell) compartment transfering B cells (or CD4 T cells)into lightly irradiated host mice. I don't know what irradiation dose I should use (400 cgy ?) and how long after irrdiation and transfer the mice will have reconstituted their lymphoid compartments and when I will be able to challenge them with Ag. What chimaerism can I expect ? (I can't use Ig or TCR transgenic donor cells).
