When we prepared the mentioned solution, we need to incubate a small portion of it at 37 degree for at least 3 days to test for any contamination. Why is it at least 3 days? What will happen if we incubate less than 3 days?

You're providing an opportunity for any bacterial contamination to grow in your solution. If it turns cloudy within the three days, you need to sterilize the solution more carefully. If you incubate less than three days, you may miss the contamination and use the contaminated solution.
