This is my first time trying to make competent cells  ;) , and i'm going to adopt the protocol from Sambrook. the question is do I have to use CaCl2 and MgCl2 with molecular biology grade? or biochemical grade is okay?  thanks before

Biochemical grade would be sufficient. I think I autoclaved my cacal2 1M.
you can autoclave your glycerol as well, but I never did.

I use invitrogen's protocol for making TOP10

biochemical grade would be sufficient i guess. I normally sterile filter my solutions. Good luck!

Thank you Curits and Biolab12 for the answers, making me more confident
And Yes, we will surely sterile filter the solutions..
