Hello everyone
Ive been trying to extract bacterial total RNA using a Qiagen RNA easy kit. Ive used a 18-20 hr grown culture for RNA extraction. I followed ll the instruction given in the manual but im not getting RNA. Pls help

Blame the RNAses.
Use RNAse inhibitors and do your best to avoid RNAses.

What cells are you using?
How do you lyse them?
Try to extract the RNA by using TRIZOL and precipitate it with isopropanol afterwards ...normally this works fine. We collect our cells in 5% phenol solution ...this prevents post-harvest degradation of RNA.
