I was wondering if anyone has advice regarding which cell lines most efficiently express transgenes following transient transfection.
I am not asking which cell lines are most efficiently transfected, but rather which cell lines express genes to the highest levels once they are transfected (although I realize that transfection efficiency and transgene expression efficiency may be related).

The efficiency of expression is usually related to the plasmid rather than the cell line (I think). Most of the commonly used cells such as 293 will transfect and express at high levels during a transient transfection.
Having said that you should use a cell type that is a model for what you are looking at e.g. A549 for lung cancer, mcf-7 for beast cancer etc.

I totally agree with Bob1. You can also use viral-transformed cell lines, such as 293T cells that express the large T antigen. They will produce a ton of protein during a transient transfection. But it will depend on the model you are looking to work with.

it depends on type of cell AND type of overexpressed protein; as suggested HEK-293 are often used...
