Hi,
My colleagues told me that because T4 buffer has ATP it is not recommended to use the buffer that has been thawed before.

They usually aliquot their buffer into 10 ul stocks when it is freshly delivered from company.

Is this really necessary? I have used previously thawed buffer and didn't have much problem.

It's not necessary. I am thawing it many times without any significant impact.

I freeze and taw my buffer always untill I run out of T4, never had problems with it.
(however, I do not use it so many times till its all used, so not sure if this counts)

On the other hand, you might wanne check this topic, and more specific HomeBrew his post.

If you don't know it, then ask it! Better to ask and look foolish to some then not ask and stay stupid.
