im not that experience in comet assay but i am very interested and very much willing to learn. but since there's no one here in our laboratory section knows how the procedure goes, i just tried the method of Singh NP, Tice RR, et al. and so far i saw the comet in my slides.  

now i would like to try the use of gamma radiation exposure on my blood samples (lymphocytes). can anyone pls help me on how to isolate lymphocytes from the whole blood. Im actually citing the study of Vera Garaj-Vrhovac, Davor Zeljezic in "Comet assay in assessment of the human genome damage induced by y-radiation in vitro".

im also asking favor if anyone knows any simple and detail protocol on the said topic for me to experienced the differences of comet tail (DNA damage) from the healthy blood (lymphocytes) to irradiated one.
