Hi
I am trying to overexpress a gene in zebrafish for which I have injected in-vitro transcribed RNA in the embro at 2 cell stage.
the RNA quality was checked on the gel before injection and I saw intact transcript there. As a control, I have also injected GFP RNA.
at 24 hpf onwards, I can see GFP expression but my gene of interest doesnt seem to be expressing at all( I have checked the expression with western blot). I dont see any reason for this as i have cloned d entire CDS (from ATG to to TGA).
Can anyone tell me the possible reason and solution for this problem???
