Hi all,
I'm having trouble sterile filtering a 200 mg/ml solution of bovine fibrinogen solution. It simply won't go through a .22 uM filter. I've tried to "step down" through several filters from larger to smaller filter sizes, without success.
Sigma says the fibrinogen solution needs to be dissolved in 0.85-.9% NaCl. I've used both DMEM (.64% NaCl) and F10 (.74% NaCl) to no avail. The fibrinogen solution takes about 4 hours at 37 degrees C to dissolve. The solution is clear after 4 hours, so it's clearly dissolving to some extent, just not enough.
Any suggestions? This problem is killing this project. Thanks.
